چکیده

Hypoxia to the tissue plays a significant role in ischemia. In spite of the development of various in vitro models of neuronal cultures to study the mechanism of ischemia-induced cell death, not much is known about the role of protein expression in ischemia induced pheochromocytoma (PC12) cells. Present study was done to study the expressions of proteins in hypoxia induced PC12 cells and after induction with conditioned media of human neuroblastoma (IMR32) and human glioblastoma (U87MG). We found down regulation of four protein markers with molecular weight of 97, 75, 40 and 14 kDa in hypoxic PC12 cells. Majority of these proteins reappeared after treatment with the conditioned media of IMR32 and U87MG cell lines. Identified proteins will be helpful to understand the mechanism of hypoxia/ischemia induced cell death and also the neuroprotective role of conditioned medium of IMR32 and U87MG cell at the proteomic level.

Secretory factors of human neuroblastoma (IMR-32) and human glioblastoma (U87MG) cell lines induce neurite outgrowths in PC12 cells.

Neurite outgrowth is essential for the communication of the nervous system. The rat Pheochromocytoma (PC12) cells are commonly used in the neuronal cell study. It is well known that exogenous stimuli such as Nerve Growth Factor (NGF) induce neurite outgrowth. In the present study it has been investigated whether or not the conditioned medium from human neuroblastoma cell line (IMR-32) and human...

Morphine-induced apoptosis in PC12 cells: role of Bax and Bcl2

Introduction: It was reported that morphine could induce apoptosis in neurons. However, its specific mechanistic pathways remain elusive. The present study was undertaken to determine whether morphine could induce apoptosis in PC12 cells, a neuronal cell line, and the involvement of Bax and Bcl-2, as upstream factors of mitochondrial pathway. Methods: In an experimental study, the viabili...

Neuroprotective effect of Nigella sativa hydro alcoholic extract on serum/glucose deprivation induced PC12 cells death

Introduction: The Serum/Glucose deprivation -induced cell death in cultured PC12 cells represents a useful in vitro model for the study of brain ischemia and neurodegenerative disorders. Nigella sativa L. has been known as a source of antioxidants. To elucidate the neuroprotective actions of N. sativa extract in vitro, we studied the effect of N. sativa extract on cultured PC12 cells under s...

Antioxidant Potential of Fagonia arabica Against Chemical Ischemia Induced in PC12 Cells

The imbalance between pro-oxidants and anti-oxidants leads to generation of oxygen/nitrogen free radicals which are implicated in several neurodegenerative diseases. Fagonia arabica is an ethno-pharmacologically important Ayurvedic herb known to have many medicinal properties like anti-inflammatory, analgesic and antipyretic effects. However, its antioxidant potential has not been investigated ...

Treatment with conditioned culture media with human mesenchymal stem cells on reserpine-induced thermal fibromyalgia model in rats

Background and Aims: Fibromyalgia (FM) is a chronic pain syndrome affected by the hippocampal function. Mesenchymal stem cells-conditioned medium (MSC-CMs) has shown antioxidant and neuroprotective potentials in treating various diseases. The present study aimed to investigate the efficacy of MSC-CMs on the thermal treatment of reserpine-induced FM and evaluate the concentration changes of tumo...
